Lipid-Based Nanoparticles for Delivery of Vaccine Adjuvants and Antigens: Toward Multicomponent Vaccines

نویسندگان

چکیده

Despite the many advances that have occurred in field of vaccine adjuvants, there are still unmet needs may enable development vaccines suitable for more challenging pathogens (e.g., HIV and tuberculosis) cancer vaccines. Liposomes already been shown to be highly effective as adjuvant/delivery systems due their versatility likely will find further uses this space. The broad potential lipid-based delivery is highlighted by recent approval COVID-19 comprising lipid nanoparticles with encapsulated mRNA. This review provides an overview different approaches can evaluated design protein, carbohydrate, nucleic acid-based antigens how these strategies might combined develop multicomponent

Micelle-Based Adjuvants for Subunit Vaccine Delivery

In the development of subunit vaccines with purified or recombinant antigens for cancer and infectious diseases, the design of improved and safe adjuvants able to efficiently target the antigen presenting cells, such as dendritic cells, represents a crucial challenge. Nanoparticle-based antigen delivery systems have been identified as an innovative strategy to improve the efficacy of subunit va...

Biodegradable Nanoparticles as Vaccine Adjuvants and Delivery Systems: Regulation of Immune Responses by Nanoparticle-Based Vaccine

Polymeric nanoand microparticles have recently been shown to possess significant potential as drug delivery systems. In particular, the use of biodegradable polymeric nanoparticles with entrapped antigens such as proteins, peptides, or DNA represents an exciting approach for controlling the release of vaccine antigens and optimizing the desired immune response via selective targeting of the ant...

Liposome-based Co-delivery of Adjuvants and Peptide Antigens: Application to the Development of Synthetic Vaccines

Introduction: Subunit vaccines represent an important aspect of modern vaccination. In this respect, the possibility to construct peptide-based vaccines is particularly appealing and peptides that e.g. mimic epitopes of pathogens, can be designed to induce well-defined monofunctional immune responses. Multi-epitopic constructs that contain epitopes from the same and/or different pathogen(s) can...

Polyethylenimine-based micro/nanoparticles as vaccine adjuvants

Vaccines have shown great success in treating and preventing tumors and infections, while adjuvants are always demanded to ensure potent immune responses. Polyethylenimine (PEI), as one of the well-studied cationic polymers, has been used as a transfection reagent for decades. However, increasing evidence has shown that PEI-based particles are also capable of acting as adjuvants. In this paper,...

Beyond antigens and adjuvants: formulating future vaccines.

The need to optimize vaccine potency while minimizing toxicity in healthy recipients has motivated studies of the formulation of vaccines to control how, when, and where antigens and adjuvants encounter immune cells and other cells/tissues following administration. An effective subunit vaccine must traffic to lymph nodes (LNs), activate both the innate and adaptive arms of the immune system, an...
